Estes The major barrier to research and development of effective interventions for human noroviruses (HuNoVs) has been the lack of a robust and reproducible in vitro cultivation system. HuNoVs are the leading cause of gastroenteritis worldwide. We report successful cultivation of multiple HuNoV strains in enterocytes in stem cell–derived, nontransformed human intestinal enteroid monolayer cultures. Bile, a critical factor of the intestinal milieu, is required for strain-dependent HuNoV replication. Lack of appropriate histoblood group antigen expression in intestinal cells restricts virus replication, and infectivity is abrogated by inactivation (e.g., irradiation, heating) and serum neutralization. This culture system recapitulates the human intestinal epithelium, permits human host-pathogen studies of previously noncultivatable pathogens, and allows the assessment of methods to prevent and treat HuNoV infections. http://science.sciencemag.org/co ... /24/science.aaf5211 |

rojjer 发表于 2016-8-30 12:11 HBV只能在人原代肝细胞培养复制,限制太大。2012年nibs的李文辉教授发现了乙肝受体NTCP,现在向HepG2稳转NTCP就可以感染HBV了。 HCV只有JFH-1这一株病毒能在体外肝细胞如Huh-7等中培养。在1999年日本科学家Wakita从一位病人中分离得到了JFH-1之一2a亚型的HCV,后来对其他几型HCV的研究都是通过替换JFH-1的结构蛋白的复制子来进行。 |
